Effects of Various Amount Levels of Chitosan Oligomer on the Disorders of Hepatic Functions, Antioxidant Enzyme Activities and Immune Related Factors in Rats Treated with 2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD)

This study was conducted to investigate the effects of various amount levels of chitosan oligomer on the disorders of hepatic functions, antioxidant enzyme activities and immune related factors induced by 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD), that is an endocrine disrupter, using adult rats for 3 weeks. The 40 SD (male, 7 weeks old)rats were divided four groups. Chitosan oligomer (MW, 5000-10000) was added as 3% or 5% levels to basal diets, respectively. TCDD (40 ug/kg B.W) was injected intraperitoneally to all rats without control, at the beginning of experiment. The amount of daily weight gain and food intake, and food efficiency ratios were significantly decreased by treating TCDD. Relative weights of thymus were apparently decreased, by the way, those of liver were markedly increased, in rats treated TCDD. The levels of white blood cells (WBC) and platelets were significantly lowered by treating TCDD, however, WBC were recovered by chitosan oligomer diets. In hepatic function, the elevation of alkaline phosphatase activities by TCDD treatment was diminuated by chitosan oligomer diets. The suppression of superoxide dismutase activities induced by treating TCDD was significantly recovered according to adding chitosan oligomer. In hepatic microsomal cytochrome p(CYP)-450, NADPH CYP-450 reductase, ethoxycoumarin-o-deethylase and benzphetamine N-demethylase, 5% chitosan oligomer diets apparently decreased these increased levels by TCDD treatment. In histochemical observation, the fat droplets and apoptosis of hepatocytes by TCDD treatment were markedly alleviated by chitosan oligomer diets. In immunoglobulin(Ig) production, the levels of IgE in spleen were significantly higher than control by chitosan oligomer diets. These results indicate that chitosan oligomer can exert improving effects on some disorders of hepatic functions, super- oxide dismutase activities and immune related factors, caused by TCDD.
